Trypsin-EDTA Solution (1X), 0.25% Trypsin (1:250) and 1 mM EDTA in Hank's Balanced Salt Solution. Without Calcium and Magnesium. Porcine Parvovirus and Mycoplasma Tested. Shipped frozen, is used in tissue culture to detach adherent mammalian cells from the culture vessel by breaking down cell-cell and cell-substrate interactions. Trypsin is a proteolytic enzyme that cleaves peptide bonds, while EDTA inactivates calcium and magnesium ions needed for cell adhesion. This process aids in passaging, subculturing, and harvesting cells for downstream assays. Optimal concentration and exposure time must be determined for different cell types to maintain cell viability. After detachment, trypsin activity from Trypsin-EDTA Solution (1X) is halted by adding a medium and serum combination to protect the cells.
