How to Prepare 125 mL Roots™ Media
- Gather Materials
- 1 sachet Roots™ media (makes 125 mL)
- 125 mL distilled or RO water
- 0.5 g Supreme Tissue Culture Agar
- pH meter or pH paper
- Magnetic stirrer or sterile stir rod
- Autoclavable flask or bottle (for mixing)
- Culture vessels (jars, tubes, or containers)
- Autoclave or Pressure cooker - Dissolve the Media
- In an autoclavable flask, add ~100 mL of - distilled/RO water.
- Pour the entire 125 mL Roots™ sachet into the water.
- Add 0.5 g agar.
- Stir gently until the powder is fully dissolved.
- Add the remaining water to bring the total volume to 125 mL. - Adjust pH
- Measure the pH.
- Adjust to 5.7–5.8 using small drops of 1 N NaOH (to raise) or 1 N HCl (to lower). Alternately, use hydroponics pH up and pH down. - Prepare for Sterilization
- Loosely cap the flask with the prepared media.
- Place your empty culture vessels (jars, tubes, or containers) in the autoclave as well, with loose-fitting lids to allow steam to enter. - Autoclave
- Autoclave the media flask and the empty vessels together at 121 °C (15 psi) for 15–20 minutes. - Pour Media into Sterile Vessels
- After autoclaving, allow the media to cool to about 50–55 °C—still liquid but not too hot.
- In a sterile laminar flow hood, carefully remove the sterilized vessels.
- Pour the cooled, sterile media into the sterile vessels.
- Let the media fully cool and solidify inside the vessels.
- Close the vessels with their sterilized lids. - Inoculate Explants
- Once the media is completely cooled and solidified, place your explants (Stage III rooting) into the vessels under sterile conditions.
- Seal the containers and move them to the proper culture environment.
Key Notes:
- Vessels and media are sterilized in the same autoclave run, saving time.
- Always pour inside a sterile laminar flow hood to avoid contamination.
- Never put explants into warm media.
- Label vessels with date, medium type, and plant name.
