Solidifying agents play a vital role in tissue culture media, providing essential support for the optimal growth and development of plants when incorporated into the medium. Additionally, the inclusion of solidifying agents helps ensure the even distribution of nutrients, hormones, and other crucial elements throughout the culture medium.
The two most common solidifying agents used in the process are agar and gellan gum. Agar, derived from seaweed, is a polysaccharide. It imparts a gel-like texture to the medium, facilitating the adherence, growth, and differentiation of plant cells.

How To Use Agar
Follow the given steps to prepare 1 liter of tissue culture media using PCT’s Supreme TC Agar:
- Take a beaker and add 800 ml of distilled water to it.
- To the water add 4.54 grams of PCT’s MS Media (or alternative medium based on your plant), 30 grams of sugar, and PPM™ (generally 1-2 ml/L).
- Make up the volume of the media to 1000ml.
- Adjust the pH of the media between 5.4-5.8.
- Add 6-8 grams of PCT’s Supreme Tissue Culture Grade Agar to the media and keep stirring.
- Autoclave the medium for 20 minutes at 15 psi and 121℃.

I am a hobbyist in Plant tissue culture, I have found this agar to be a great consistency and easy to use. I had very fast shipping to New Zealand .
I originally receive the small size of agar and as soon as I was half way through the small bottle I just had to order a KG! This stuff works great for most cannabis cultivars and works great for other plant species too. Definitely worth it! I throw in a silica gel pack in so no humidity builds up in the bottle after opening and closing, this keeps it fresh with no clumping. I’ll be testing this for mycology next, I’m certain it will work great for that too! I’ve tried other companies and others have not worked as good as this. Definitely in my top 2 preferred gelling agents! Will definitely buy again! Probably even a bigger size next time! I’ll be testing it on avocados and other tropical plants next. I have been using the PCT Supreme Grade Agar for plant tissue culture and orchid seed germination for several months and have found it to be very good in terms of performance, color (essentially colorless), clarity, and gel strength. At about 6.5 g/L it gives me a very good gel for my purposes. The price is very good too.
