Using PPM to Determine Sterilization Rates During Tissue Culture

"On the first round of experiments, I used 4% PPM (10 ml. 9.6 ml of sterilize water and 0.4 ml of PPM) for 4 hours. The results weren't great, with only 4 out of the 20 plants surviving - a majority were killed during the sterilization process. While the results may look negative, I'm actually pretty happy with the results.
Yes, some of the plants were killed, but of the ones that survived no contamination was present! That means I just need to reduce the sterilization time until I found the "sweet spot." It's important to note that these plants are very sensitive, so it's normal to run tests to determine the right times and dosages for sterilization.
For those plants that did get contaminated, I'm going to try the same percentage of PPM and time, but I'm going to make sure my explants are fully clean with no soil or anything that may cause contamination problems."
