Murashige and Skoog Media, MS Media is a nutrient media used for the in vitro culture of plant cells, tissues, and organs. It was first developed in 1962 by Toshio Murashige and Folke Skoog and has since become one of the most widely used media for plant tissue culture.

It contains a balanced mixture of macronutrients, micronutrients, vitamins, and plant growth regulators to provide optimal conditions for the proper growth and development of plants.

The Benefits and Applications of MS Media

  • Easy to use
  • Supply nutrients required by in vitro plants for their proper growth and development
  • Provides a well-balanced composition of nutrients, including essential minerals and vitamins, supporting the growth of various plant tissues.
  • Effective in inducing callus formation
  • A dynamic supplier of numerous amino acids, organic acids, plant growth regulators (such as cytokinins and auxins), vitamins, sugars, minerals, nucleic acids, and unidentified growth-promoting substances.

How To Use MS Media

Follow the given steps to prepare 1 liter of tissue culture media using MS media:

  1. Take a beaker and add 800 ml of distilled water to it.
  2. To the water add PCT’s MS Media 4.54 grams, 30 grams of sugar, and PPM™ (generally 1-2 ml/L)
  3. Make up the volume of the media to 1000ml.
  4. Adjust the pH of the media between 5.4-5.8.
  5. Add gelling agent (3-4 g/L gellan gum or 6-8 g/L agar) to the media and keep stirring.
  6. Autoclave the medium for 20 minutes at 15 psi and 121℃.

Your media is ready!

Note: Media should be prepared according to the formula mentioned on the label. However, it is recommended to use an entire container at once.

Heat-liable substrates should be added, after autoclaving.
